Panicked tourists on board the vehicle, who included families with young children, said afterwards they thought at first they were being attacked by terrorists.
Tourist Andrew Carey, 39, from Bridgend, South Wales, was on the top deck of the bus while on a romantic break with his wife Natalie.
He said: ‘I really thought it was a terrorist attack and my number was up.
‘Masked men surrounded the bus and began shouting. We were getting ready for someone to come up the stairs with a knife or a gun. It was a relief they just sprayed graffiti. It was very frightening.’
The men daubed the words ‘El Turisme Mata Els Barris’ in Catalan across the windscreen of the bus, which in English translates as ‘Tourism Kills Neighbourhoods.’
City hall chiefs have sought to play down the attack by describing it as an ‘isolated incident’ although they admitted it was the work of an organised group.
No-one has yet been arrested over the incident, which happened around 11am on Thursday but only came to light Sunday.
It occurred less than three months after demonstrators protesting over the tourism boom in their Barcelona neighbourhood of Poblenou threw paint and eggs at two hotels at two hotels during a protest march.
Guests at one hotel – the Travelodge Poblenou – were warded away from the reception area after a smoke canister was let off outside. The other hotel targeted was the Amistat Beach Hostel near Poblenou tube station.
The four men wrote ‘tourism kills neighbourhoods’ in Catalan on the front of the bus
Police monitoring the May 5 demonstration, organised by a neighbourhood group called Ens Plantem, said no-one was arrested but three people were identified.
Around 300 people took part in the street protest to criticise the gentrification of the area and high rental prices which have soared with the influx of tourists to an area once dubbed the Catalonian Manchester.
It fell into a state of abandon before a dramatic transformation triggered by the Olympic Games in 1992.
Ens Plantem has been fighting the construction of two new hotels planned for the area which they say has caused structural problems in neighbouring buildings, and have called for schools to be built in their place.
The incidents were described at the time as the first of their kind in the last few years of growing tension in several Barcelona neighbourhoods over the effects of mass tourism.
Tensions over tourism has increased in recent years in Barcelona.
In January 2,000 people descended on Las Ramblas in Barcelona (pictured) to protest against the massive influx of tourists
Locals in working class neighbourhood La Barceloneta have protested against drunken Magaluf-style holidaymakers and illegal tourist flats.
There have also been demonstrations around the Gaudi-designed Sagrada Familia to complain about the daily hordes of visitors.
Nearly 2,000 people demonstrated in Barcelona’s famous Las Ramblas at the start of the year over claims the tourism boom is out of control and affecting local people by pricing them out of their own neighbourhoods through higher rents and property sales.
There has also been tension in the Balearic Islands.
Vandals daubed buildings in the Majorcan capital Palma including a four-star hotel with anti-tourist messages in April last year.
There have also been demonstrations about the daily hordes of visitors to the Sagrada Familia
One on the four-star Palma Suites Aparthotel in Palma old town, written in English like most of the others, said: ‘Tourist Go Home. Refugees Welcome.’
Others carrying the same message had been daubed on walls and waste paper bins in the same area including one on a private tourism school.
‘Guiris Go Home’, the slang word the Spanish use to describe British and other northern European holidaymakers, and the more threatening slogan ‘Tourists, You Are the Terrorist.’ also left visitors with a sour taste in their mouths.
More graffiti was daubed around Palma earlier this month.
Locals in Ibiza have complained of having to share their swimming pool in the apartment blocks where they live with holidaymakers drinking and taking drugs in front of their children in broad daylight.
It is not known if any Brits or Irish tourists were on board the Barcelona bus attacked.
